Project Description

The proposed research project addresses a critical and current challenge in the conservation of animal genetic resources: the effective cryopreservation of boar semen from endangered indigenous European pig breeds. These breeds, including the Hungarian Large White Pig, the Mangalica Pig and the Slovenian Krškopolje Pig, have unique adaptive traits, excellent meat quality and cultural significance. However, they are endangered due to disease outbreaks, such as African Swine Fever, climate change, and the loss of genetic diversity. Despite advances in reproductive biotechnology, frozen semen is used in less than 1% of artificial inseminations of pigs, largely due to the inherent cryosensitivity of boar semen. This project aims to fill a significant knowledge gap by investigating the breed-specific and individual factors that influence semen freezability in these under-researched populations. The research will follow a three-phase, multidisciplinary approach. First, semen samples from commercial and indigenous boars will undergo a comprehensive quality assessment using advanced molecular, genomic and proteomic techniques. Secondly, conventional and experimental cryopreservation protocols will be applied and evaluated, focusing on biomarkers (e.g. AKAP3, FN1) and gene expression profiles (e.g. PRM1, PRM2). Finally, optimized cryopreservation protocols will be developed and validated for practical application in gene banks and breeding programs. These protocols will not only ensure the long-term ex situ conservation of valuable genetic traits, but also support sustainable pig farming by promoting genetic diversity and the rural economy. The project integrates state-of-the-art cryobiology, molecular diagnostics and conservation science. It involves close cooperation between Hungarian and Slovenian institutions and includes the active participation of young researchers. The expected results will significantly advance reproductive biotechnology and provide scalable solutions for the conservation of livestock biodiversity, thus contributing to both scientific innovation and the conservation of agrobiodiversity on a European and global scale.
